The COMT gene provides instructions for making catechol-O-methyltransferase, an enzyme involved in breaking down catechol compounds, including dopamine, norepinephrine, and epinephrine.
The best-known COMT variant is rs4680, also called Val158Met. This common genetic difference changes the stability and activity of the COMT enzyme. Depending on your genotype, you may have the Val/Val, Val/Met, or Met/Met form.

What Does the COMT rs4680 Variant Mean?
The rs4680 variant changes one amino acid in the COMT enzyme [R]:
the G allele encodes valine, called Val;
the A allele encodes methionine, called Met.
The Met form is less stable and is associated with lower COMT enzyme activity. The Val form is more stable and has higher activity relative to the Met form. Functional research found that rs4680 affects COMT protein abundance and enzyme activity without producing the same clear difference in messenger RNA expression [R].

What Does the COMT Gene Do?
The COMT gene is located on chromosome 22 and encodes the catechol-O-methyltransferase enzyme [R].
COMT transfers a methyl group from S-adenosylmethionine to catechol compounds [R]. This reaction contributes to the metabolism of:
dopamine;
norepinephrine;
epinephrine;
catechol estrogens;
certain medications and other catechol-containing substances.
COMT is produced in two principal forms: soluble COMT and membrane-bound COMT. The relative importance of these forms differs between tissues [R]. COMT is only one part of catecholamine metabolism. Other enzymes, transporters, receptors, and regulatory processes also influence how dopamine and related signaling systems function.

What Is COMT rs4680?
The rs4680 variant is a common missense variant in the COMT gene. It changes the amino acid at position 158 of membrane-bound COMT from valine to methionine, which is why it is called Val158Met [R]. The same variant may also be described as Val108Met when referring to the shorter soluble form of the enzyme. The notation Val108/158Met refers to the same rs4680 variant in the two COMT protein forms [R].
Research using postmortem human brain tissue found genotype-related differences in COMT protein abundance and enzyme activity, with [R]:
Val/Val showing the highest activity;
Val/Met showing intermediate activity;
Met/Met showing the lowest activity.
How Does COMT Relate to Dopamine?
COMT participates in the breakdown of dopamine, but its importance is not identical throughout the brain and body. Researchers have been particularly interested in COMT’s role in the prefrontal cortex, a region involved in working memory, attention, decision-making, and other executive functions [R]. Differences in COMT activity may influence aspects of dopamine signaling in this region.
The Met allele’s lower enzyme activity has therefore sometimes been interpreted as supporting greater prefrontal dopamine availability, while the Val allele’s higher activity has been interpreted as supporting faster dopamine breakdown. That explanation is useful for understanding the proposed mechanism, but it is not a direct measurement of dopamine.
Your rs4680 genotype cannot determine:
your current dopamine level;
dopamine activity throughout the entire brain;
whether your dopamine is “too high” or “too low”;
whether you have a dopamine-related medical condition;
how you will respond to every stressful or rewarding situation.
Dopamine signaling also depends on its production, release, transport, receptor activity, metabolism through other pathways, brain region, health status, medications, and environmental factors [R].
Does COMT Determine Personality?
No. A COMT genotype does not determine personality.
Val/Val is sometimes described online as the “warrior” genotype, while Met/Met is called the “worrier” genotype. These labels reduce complicated and sometimes inconsistent research findings to personality categories that cannot reliably describe an individual.

Some individual studies and neuroimaging analyses have reported genotype-related differences. However, these findings depend on the population, task, environment, sex, age, health status, and other variables.
Is rs4680 a COMT Mutation?
Rs4680 is better described as a common functional genetic variant or polymorphism [R]. The word “mutation” is sometimes used informally for any difference in DNA, but it can misleadingly suggest that rs4680 is a rare disease-causing variant.
Rs4680 is common in the population, and none of its three genotypes is inherently a diagnosis. The variant changes enzyme activity, but it is not equivalent to a pathogenic variant that independently establishes a genetic disorder.
Does COMT Affect Stress or Emotional Processing?
COMT has been studied in relation to stress and emotional processing because catecholamine signaling contributes to how the brain responds to cognitive and emotional demands [R, R].
Some studies have reported different patterns between Val and Met carriers under particular experimental conditions [R, R]. Other studies have failed to reproduce those associations or have found that the result changes according to sex, ancestry, age, diagnosis, or environmental exposure.
This means that rs4680 should not be used to:
diagnose anxiety, depression, ADHD, or another condition;
predict emotional resilience;
determine whether someone handles stress well;
classify someone as emotionally stable or vulnerable;
explain a psychiatric symptom by itself.
