DRD2 Gene Explained: rs6277, rs1800497, and Dopamine

The DRD2 gene provides instructions for making the dopamine D2 receptor. Researchers have studied common variants near or within this gene for possible effects on dopamine-receptor binding, reward learning, and behavior.

Two frequently discussed markers are rs6277, also called C957T, and rs1800497, often called Taq1A. The names can be confusing: rs6277 is in DRD2, while rs1800497 is in the neighboring ANKK1 gene. Both have been studied using brain-imaging measures of receptor availability, but neither tells you how much dopamine you have or predicts your personality, motivation, or addiction risk [R, R].

Quick Answer: What Do DRD2 rs6277 and rs1800497 Mean?

  • rs6277 (C957T) is a synonymous variant within DRD2. In GenesUnveiled’s forward-strand notation, the alleles are G and A; the literature’s C957T “T” allele corresponds to forward-strand A. Small PET studies have found higher striatal D2-receptor binding measures with increasing A-allele copies, but this is not a direct measurement of receptor number or dopamine [R, R, R].

  • rs1800497 (Taq1A) lies in ANKK1 next to DRD2. The A1 form is shown as A in forward-strand genotypes used by GenesUnveiled. A meta-analysis of five imaging studies (194 healthy participants) found lower striatal D2-receptor binding potential among A1 carriers than among people with no A1 copies [R].

  • The meta-analysis reported a standardized mean difference of −0.57 for rs1800497, with an estimated about 7% of variation in the imaging measure explained. That is not a 57% reduction or a fivefold difference, and it does not predict an individual’s brain function [R].

What Does the DRD2 Gene Do?

DRD2 encodes the dopamine D2 receptor, a G-protein-coupled receptor expressed in several parts of the brain and body. In the brain, D2 receptors are abundant in the striatum, a region involved in movement, learning from outcomes, and reward-related processes [R]. When dopamine binds to D2 receptors, it changes signaling inside the receiving cell. D2 signaling is one part of a broader network that includes other dopamine receptors, dopamine transporters, neurotransmitter systems, and connections among brain regions.

That context is important. A DNA variant associated with a brain-imaging measure does not directly reveal someone’s dopamine level, how strongly they feel reward, or how they will behave. The imaging studies discussed below measure receptor binding potential—not motivation, pleasure, intelligence, or addiction.

Why Are rs6277 and rs1800497 Often Discussed Together?

The two markers are close to each other on chromosome 11, but they are not the same variant and are not in the same gene:

  • rs6277 is a synonymous variant in the DRD2 coding sequence. It does not change the receptor’s amino-acid sequence.

  • rs1800497 is in exon 8 of the neighboring ANKK1 gene. It is often called “DRD2 Taq1A” because of its historical association with dopamine D2 receptor research, but it is not inside DRD2 [R, R].

Both have been investigated for associations with D2-receptor binding. That does not mean either marker alone explains differences in dopamine signaling. rs1800497 may also tag other nearby variants through linkage disequilibrium, so an observed association does not prove that rs1800497 itself causes the imaging difference [R].

DRD2 rs6277 (C957T): What the Genotypes Mean

The name C957T comes from the DRD2 transcript strand used in much of the research. On the forward genomic strand, rs6277 is G/A, so the transcript C957T “T” allele corresponds to forward-strand A. GenesUnveiled uses forward-strand alleles, so the marker appears as G/A in a raw DNA result [R, R].

  • GG corresponds to C/C in the C957T literature: no copies of the transcript T allele.

  • GA corresponds to C/T: one transcript T allele.

  • AA corresponds to T/T: two transcript T alleles.

In one PET study of 38 healthy men, measured striatal D2-receptor availability followed the pattern C/C < C/T < T/T. A later PET study of 84 people reported a positive relationship between C957T T-allele dosage and D2/3-receptor binding in parts of the striatum, including the putamen and ventral striatum. In forward-strand notation, those findings correspond to GG < GA < AA for the imaging measure [R, R].

These studies do not establish a precise percentage or fold difference for each genotype. The samples were modest, and PET binding potential is influenced by both receptor availability and tracer binding properties. One study attributed its genotype-related result in part to altered receptor affinity. It should not be translated into “more dopamine” or a predictable behavioral effect [R].

Laboratory research adds another layer. In cells, the C957T transcript T form altered predicted mRNA folding and was associated with reduced mRNA stability and translation. That result is not a simple match for the higher PET binding measure reported with T-allele copies in some human studies. The findings concern different biological measurements, and the complete mechanism remains unresolved [R].

ANKK1 rs1800497 (Taq1A): What the Genotypes Mean

For rs1800497, in forward-strand notation the A1 form is A and the A2 form is G:

  • GG is A2/A2: no A1 copies. This was the comparison group in the main receptor-availability meta-analysis.

  • GA is A1/A2: one A1 copy.

  • AA is A1/A1: two A1 copies.

The research synthesis grouped GA and AA together as A1 carriers and compared them with GG. It did not establish that AA has exactly twice the effect of GA [R, R].

In a meta-analysis of five molecular-imaging studies with 194 healthy participants, A1 carriers had lower striatal D2-receptor binding potential than A2/A2 participants. The weighted standardized mean difference was −0.57 (95% CI −0.87 to −0.27). The authors estimated that rs1800497 explained about 7% of the observed variation in this imaging measure in those samples (95% CI 1.7%–15%) [R].

rs1800497 changes the ANKK1 protein rather than the amino-acid sequence of the D2 receptor. It is not yet clear whether rs1800497 itself is the cause of the receptor-binding association or whether it marks another nearby variant [R, R].

Do DRD2 Variants Predict Addiction or Alcohol Use Disorder?

No. rs6277 and rs1800497 are not diagnostic tests for addiction, and neither can determine whether someone will develop an alcohol or substance use disorder.

The rs1800497 association with alcohol dependence has been debated for decades. A 2019 meta-analysis of 62 studies and 16,294 participants reported a pooled odds ratio of 1.23. However, the authors found that the association was driven by unusually low A1-allele frequencies in control groups in positive studies. Their analyses did not support rs1800497 itself as a variant that alters DRD2 expression, and they did not find significant associations for DRD2-region variants in the three clinical samples they examined [R].

Do These Genotypes Tell You How Much Dopamine You Have?

No. A raw DNA result cannot measure current dopamine concentration or activity in a specific brain region.

For rs6277, some PET studies report genotype-related differences in D2-receptor binding measures, while cell studies describe effects on DRD2 mRNA stability and translation. For rs1800497, a small meta-analysis found a group-level difference in striatal binding potential among healthy participants. Neither result measures dopamine concentration or establishes how a person feels, learns, or responds to reward.

Other genes, brain circuits, age, health, medications, environment, and experience also influence dopamine-related signaling. A genotype is one piece of a much larger biological picture.

What Your Raw DNA Result Can—and Cannot—Tell You

If your raw DNA file includes a readable genotype for rs6277 or rs1800497, it can show which alleles were recorded at that marker. It cannot directly measure receptor binding, dopamine levels, reward sensitivity, motivation, or addiction risk.

Allele orientation is important. Research papers may use transcript-strand names such as C957T or allele labels such as A1/A2, while GenesUnveiled displays forward-strand genotype letters. For these two markers, GenesUnveiled reports rs6277 and rs1800497 as A/G genotypes. Avoid converting a genotype by eye unless the strand orientation has been checked.

GenesUnveiled GeneExplorer provides educational interpretations of selected markers found in supported raw DNA files. It is not a diagnostic test or a substitute for clinical evaluation.

Frequently Asked Questions

Is rs1800497 in the DRD2 gene?

No. rs1800497 is in exon 8 of the neighboring ANKK1 gene. It is often called DRD2 Taq1A because it has been studied in relation to D2-receptor binding, but it is not located within DRD2.

What does rs6277 AA mean?

In forward-strand notation, rs6277 AA corresponds to two copies of the transcript T allele in the C957T literature. Some PET studies have reported higher striatal D2-receptor binding measures with more T copies, but this does not mean higher dopamine or predict a specific behavior.

What does rs1800497 AA mean?

In GenesUnveiled’s forward-strand notation, rs1800497 AA corresponds to A1/A1. A meta-analysis grouped AA and GA together as A1 carriers; it did not establish a separate exact multiplier for AA compared with GA.

Does the rs1800497 A1 allele mean lower dopamine?

No. A1 carriers had lower striatal D2-receptor binding potential in a small meta-analysis of healthy participants. Binding potential is an indirect measure, not a dopamine concentration, and it does not determine a person’s behavior or mental health.

Does the DRD2 gene determine motivation or personality?

No. DRD2 participates in dopamine signaling, but these common variants do not define personality, motivation, intelligence, or learning ability.
